چکیده

Coherent Raman scattering microscopy is an attractive new technology for label-free imaging. Both coherent anti-Stokes (CARS) and stimulated (SRS) offer the possibility to record hyperspectral imaging data. While analysis of spontaneous data multivariate methods are nowadays routinely employed, until date most interpreted using univariate analysis. In this work, we report a quantitative comparison performance different used SRS from model samples. Our show all samples that outperform Using metrics quantify method performance, find tested, curve resolution (MCR) gives best results. We combination with selection essential components based on first-order autocorrelation, simple workflow MCR-based

Quantitative vibrational imaging by hyperspectral stimulated Raman scattering microscopy and multivariate curve resolution analysis.

Spectroscopic imaging has been an increasingly critical approach for unveiling specific molecules in biological environments. Toward this goal, we demonstrate hyperspectral stimulated Raman loss (SRL) imaging by intrapulse spectral scanning through a femtosecond pulse shaper. The hyperspectral stack of SRL images is further analyzed by a multivariate curve resolution (MCR) method to reconstruct...

A machine learning framework to analyze hyperspectral stimulated Raman scattering microscopy images of expressed human meibum.

We develop and discuss a methodology for batch-level analysis of hyperspectral stimulated Raman scattering (hsSRS) data sets of human meibum in the CH-stretching vibrational range. The analysis consists of two steps. The first step uses a training set (n=19) to determine chemically meaningful reference spectra that jointly constitute a basis set for the sample. This procedure makes use of batch...

Multicolor stimulated Raman scattering microscopy

Stimulated Raman scattering (SRS) microscopy has opened up a wide range of biochemical imaging applications by probing a particular Raman-active molecule vibrational mode in the specimen. However, the original implementation with picosecond pulse excitation can only realize rapid chemical mapping with a single Raman band. Here we present a novel SRS microscopic technique using a grating-based p...

Label-free quantitative imaging of cholesterol in intact tissues by hyperspectral stimulated Raman scattering microscopy.

A finger on the pulse: Current molecular analysis of cells and tissues routinely relies on separation, enrichment, and subsequent measurements by various assays. Now, a platform of hyperspectral stimulated Raman scattering microscopy has been developed for the fast, quantitative, and label-free imaging of biomolecules in intact tissues using spectroscopic fingerprints as the contrast mechanism.

In Vivo Metabolic Fingerprinting of Neutral Lipids with Hyperspectral Stimulated Raman Scattering Microscopy

Metabolic fingerprinting provides valuable information on the physiopathological states of cells and tissues. Traditional imaging mass spectrometry and magnetic resonance imaging are unable to probe the spatial-temporal dynamics of metabolites at the subcellular level due to either lack of spatial resolution or inability to perform live cell imaging. Here we report a complementary metabolic ima...
